适应症
本品用于治疗对氯喹敏感的恶性疟、间日疟及三日疟。并可用于疟疾症状的抑制性预防。也可用于治疗肠外阿米巴病、结缔组织病、光敏感性疾病(如日晒红斑)等。
用法用量
1.成人常用量：|(1)间日疟，口服首剂1g，第2、3日各0.75g。|(2)抑制性预防疟疾，口服每周1次，每次0.5g。|(3)肠外阿米巴病，口服每日1g，连服2日后改为每日0.5g，总疗程为3周。|(4)类风湿性关节炎，每日0.25~0.5g，待症状控制后，改为0.125g，一日2~3次，需服用6周~6个月才能达到最大的疗效，可作为水杨酸制剂及递减肾上腺皮质激素时的辅助药物。|2.小儿常用量：|(1)间日疟，口服首次剂量按体重10mg/kg（以氯喹计算，以下同），最大量不超过600mg，6小时后按体重5mg/kg再服1次，第2、3日每日按体重5mg/kg。|(2)肠外阿米巴病，每日按体重口服10mg/kg（最大量不超过600mg），分2~3次服，连服2周，休息1周后，可重复一疗程。
不良反应
1. 本品用于治疗疟疾时，不良反应较少，口服一般可能出现的反应有:头晕、头痛、眼花、食欲减退、恶心、呕吐、腹痛、腹泻、皮肤瘙痒、皮疹、甚至剥脱性皮炎、耳鸣、烦躁等。反应大多较轻，停药后可自行消失。 2. 在治疗肺吸虫病、华支睾吸虫病及结缔组织疾病时，用药量大，疗程长，可能会有较重的反应，常见者为对眼的毒性，因氯喹可由泪腺分泌，并由角膜吸收，在角膜上出现弥漫性白色颗粒，停药后可消失。 3. 本品相当部分在组织内蓄积，久服可致视网膜轻度水肿和色素聚集，出现暗点，影响视力，常为不可逆。 4. 氯喹还可损害听力，妊娠妇女大量服用可造成小儿先天性耳聋，智力迟钝、脑积水、四肢缺陷等。 5. 氯喹偶可引起窦房结的抑制，导致心律失常、休克，严重时可发生阿-斯综合征，而导致死亡。 6. 本品尚可导致药物性精神病、白细胞减少、紫瘢、皮疹、皮炎，光敏性皮炎乃至剥脱性皮炎、牛皮癣、毛发变白、脱毛、神经肌肉痛、轻度短暂头痛等。 7. 溶血、再障、可逆性粒细胞缺乏症、血小板减少等较为罕见。
禁忌
孕妇禁用。
注意事项
1. 肝肾功能不全，心脏病，重型多型红斑，血卟啉病，牛皮癣及精神病患者慎用。 2. 本品可引起胎儿脑积水，四肢畸形及耳聋、故孕妇禁用。 3. 耐氯喹者效果不佳。
