适应症
本品主要适用于慢性粒细胞白血病的慢性期、对缺乏费城染色体Ph1病人效果不佳。也可用于治疗原发性血小板增多症、真性红细胞增多症等慢性骨髓增殖性疾病。
用法用量
本品成人常用量：慢性粒细胞白血病：|1.每日总量4～6mg/m2,每日一次。如白细胞数下降至20×109/L则需酌情停药。|2.或给维持量每日或隔日1～2mg，以维持白细胞计数在10×109/L左右。
不良反应
可产生骨髓抑制。常见为粒细胞减少，血小板减少。严重者需及时停药。长期服用或用药过大可致肺纤维化。可有皮肤色素沉着，高尿酸血症及性功能减退，男性乳房女性化，睾丸萎缩，女性月经不调等。白内障，多型红斑皮疹，结节性多动脉炎为罕见不良反应。曾有个别报道使用高剂量后出现癫痫发作;心内膜纤维化，并由此出现相应症状;以及少见的肝静脉闭锁。
禁忌
本品有可能增加胎儿死亡及先天畸形的危险，因此在妊娠初期三个月内不能用此药。既往对此药过敏的病人。
注意事项
1. 慢粒白血病病人治疗时有大量细胞破坏、血及尿中尿酸水平可明显升高、严重时可产生尿酸肾病。2. 对有骨髓抑制、感染、有细胞毒药物或放疗史的病人也应慎用。3. 治疗前及治疗中应严密观察血象及肝肾功的变化、及时调整剂量、特别注意检查血尿素氮、内生肌酐清除率、胆红素、丙氨酸转移酶ALT(SGPT)及血清尿酸。4. 服药应根据患者对药物的反应、骨髓抑制程度、个体差异而调整剂量。5. 嘱病人多摄入液体并碱化尿液或服用别嘌呤醇以防止高尿酸血症及尿酸性肾病的产生。6. 发现粒细胞或血小板迅速大幅度下降时应立即停药或减量以防止出现严重骨髓抑制。
