适应症
主要用于治疗抗惊厥、癫痫，是治疗癫痫持续状态的重要药物。可用于麻醉前用药。
用法用量
肌内注射：成人常用量：催眠，一次50～100mg；麻醉前用药，一次100～200mg；术后应用，一次100～200mg，必要时重复，24小时内总量可达400mg；极量一次250mg，一日500mg。治疗癫痫持续状态时剂量加大，静脉注射一次200～300mg（速度不超过每分钟60mg），必要时6小时重复一次。小儿常用量：镇静或麻醉前应用，一次按体重2mg/kg；抗惊厥或催眠每次按体重3～5mg/kg或按体表面积125mg/m2。
不良反应
1．用于抗癫痫时最常见的不良反应为镇静，但随着疗程的持续，其镇静作用逐渐变得不明显，2．可能引起微妙的情感变化，出现认知和记忆的缺损。3．长期用药，偶见叶酸缺乏和低钙血症。4．罕见巨幼红细胞性贫血和骨软化。5．大剂量时可产生眼球震颤、共济失调和严重的呼吸抑制。6．用本品的患者中约1%～3%的人出现皮肤反应，多见者为各种皮疹以及哮喘，严重者可出现剥脱性皮炎和多形红斑（或Stevens-Johnson综合症），中毒性表皮坏死极为罕见。7．有报道用药者出现肝炎和肝功能紊乱。8．长时间使用可发生药物依赖，停药后易发生停药综合症。
禁忌
禁用于以下情况：严重肺功能不全、肝硬化、血卟啉病史、贫血、哮喘史、未控制的糖尿病、过敏等。
注意事项
1．对一种巴比妥过敏者，可能对本品过敏。2．作抗癫痫药应用时，可能需10～30天才能达到最大效果，需按体重计算药量，如有可能应定期测定血药浓度，以达最大疗效。3．肝功能不全者，用量应从小量开始。4．长期用药可产生耐药性。5．长期用药可产生精神或躯体的药物依赖性，停药需逐渐减量，以免引起撤药症状。6．与其他中枢抑制药合用，对中枢产生协同抑制作用，应注意。7．下列情况慎用：轻微脑功能障碍(MBD)症、低血压、高血压、贫血、甲状腺功能低下、肾上腺功能减退、心肝肾功能损害、高空作业、驾驶员、精细和危险工种作业者。
