适应症
本品主要用于治疗焦虑、失眠(用于睡眠时间短早醒患者)?癫痫及运动障碍。是治疗癫痫大发作及局限性发作的重要药物。也可用作抗高胆红素血症药及麻醉前用药。
用法用量
1.成人常用量: (1)催眠，30～100mg，晚上一次顿服； (2)镇静，一次15～30mg，每日2～3次； (3)抗惊厥，每日90～180mg，可在晚上一次顿服，或每次30～60mg，每日三次；极量一次250mg，一日500mg； (4)抗高胆红素血症，一次30～60mg，每日三次。 2.小儿常用量: (1)用药应个体化，镇静，每次按体重2mg/kg，或按体表面积60mg/m2，每日2～3次； (2)抗惊厥，每次按体重3～5mg/kg；抗高胆红素血症，每次按体重5～8mg/kg，分次口服，3～7天见效。
不良反应
1.用于抗癫痫时最常见的不良反应为镇静，但随着疗程的持续，其镇静作用逐渐变得不明显。2.可能引起微妙的情感变化，出现认知和记忆的缺损。3.长期用药，偶见叶酸缺乏和低钙血症。4.罕见巨幼红细胞性贫血和骨软化。5.大剂量时可产生眼球震颤，共济失调和严重的呼吸抑制。6.用本品的患者中约1～3%的人出现皮肤反应，多见者为各种皮疹，严重者可出现剥脱性皮炎和多形红斑(或Stevens-Johnson综合症)，中毒性表皮坏死极为罕见。7.有报道用药者出现肝炎和肝功能紊乱。8.长时间使用可发生药物依赖，停药后易发生停药综合征。
禁忌
禁用于以下情况：严重肺功能不全、肝硬化、血卟啉病史、贫血、哮喘史、未控制的糖尿病、过敏等。
注意事项
1.对一种巴比妥过敏者，可能对本品过敏;2.作抗癫痫药应用时，可能需10～30天才能达到最大效果，需按体重计算药量，如有可能应定期测定血药浓度，以达最大疗效;3.肝功能不全者，用量应从小量开始;4.长期用药可产生精神或躯体的药物依赖性，停药需逐渐减量，以免引起撤药症状;5.与其他中枢抑制药合用，对中枢产生协同抑制作用，应注意;6.下列情况慎用:轻微脑功能障碍(MBD)症、低血压、高血压、贫血、甲状腺功能低下、肾上腺功能减退、心肝肾功能损害、高空作业、驾驶员、精细和危险工种作业者。
