适应症
1.尿嘧啶合用,可提高氟尿嘧啶的疗效,临床上常用于结直肠癌与胃癌的治疗。|2.作叶酸拮抗剂(如甲氨蝶呤、乙胺嘧啶或甲氧苄啶等)的解毒剂，本品临床常用于预防甲氨蝶呤过量等大剂量治疗后所引起的严重毒性作用。|3.当口服叶酸疗效不佳时,用于口炎性腹泻、营养不良、妊娠期或婴儿期引起的巨幼细胞性贫血。
用法用量
口服给药:|1.作为甲氨喋呤的"解救"疗法，一般采用的剂量为5～15mg，每6～8小时一次，连续2日，根据血药浓度测定结果控制甲氨喋呤血药浓度在5×10-8mol/L以下。|2.作为乙胺嘧啶或甲氧苄啶等的解毒，每日剂量5～15mg，视中毒情况而定。|3.用于巨幼细胞贫血，每日15mg。|4.与5-氟尿嘧啶合用时，20～30mg/m2体表面积，在5-氟尿嘧啶用药半小时后服。
不良反应
很少见，偶见皮疹、荨麻疹或哮喘等过敏反应。
禁忌
禁用于恶性贫血或维生素B12缺乏所引起的巨幼细胞贫血。
注意事项
1.初次使用本品，应在有经验医师指导下用药。|2.本品不宜与叶酸拮抗剂（如甲氨喋呤）同时使用，以免影响后者的治疗作用，应于大剂量使用甲氨喋呤24～28小时后应用本品。|3.当患者有下列情况者，本品应慎用于甲氨喋呤的\"解毒\"治疗：酸性尿（pH<7）?腹水、失水、胃肠道梗阻、胸腔渗液或肾功能障碍。有上述情况时，甲氨喋呤毒性较显著，且不易从体内排出，病情急需者，本品剂量要加大。|4.接受大剂量甲氨喋呤而用本品\"解救\"者应进行下列各种实验室监测:|(1)治疗前观察肌酐廓清试验。|(2)甲氨喋呤大剂量后每12～24小时测定血浆或血清甲氨喋呤浓度，以调整本品剂量;当甲氨喋呤浓度低于510-8mol/L时，可以停止实验室监察。|(3)甲氨喋呤治疗前及以后每24小时测定血清肌酐量，用药后24小时肌酐量大于治疗前50%，指示有严重肾毒性，要慎重处理。|(4)甲氨喋呤用药前和用药后每6小时应监察尿液酸度，要求尿液pH保持在7以上，必要时用碳酸氢钠和水化治疗(每日补液量在3000ml/m2)。
贮藏
遮光，严封，置阴凉处保存。
