适应症
本品适用于单独采取控制饮食无效的非胰岛素依赖性糖尿病。
用法用量
1.口服。开始时，早餐前1次，或早餐及午餐前各服1次，每次1.75 mg（1粒）。
2.必要时，可在医生的观察下每日服5.25～7 mg（3～4粒），最大用量每日不超过10.5mg (6粒)。
不良反应
1.低血糖反应:
(1)本品为磺酰脲类药物，所有磺酰脲类药物都能产生严重的低血糖，重要的是选择适当的病人和剂量以避免低血糖发生。肝肾功能不全可使格列本脲的血药浓度升高，可降低糖原异生功能，这二种情况都可以导致严重的低血糖反应。
(2)老年人、衰弱和营养不良的病人以及肾和垂体功能不足的病人特别容易产生低血糖。低血糖症在老年人和服用β-肾上腺激素阻滞剂的人很难区别。低血糖更多出现在热量摄入不足、高强度长时间锻炼、饮酒或使用多种降糖药之后。
2.胃肠道反应:很少出现黄疸和肝炎，如果出现应停止服用本品。肝功能不正常，包括转氨酶升高有报道。胃肠失调、恶心、上腹胀满和胃灼热是最常见的反应，在临床试验期间占受试病人1.8%，一般与剂量有关，剂量减少后该症状消失。
3.皮肤反应:皮肤过敏反应，瘙痒、红斑、荨麻疹和丘疹的出现在临床期间占1.5%，继续服用本品，可能消失，如皮肤过敏持续不消失，应停止服用本品。另有报道，服用磺酰脲类降糖药有皮肤吡咯紫沉着症和光敏反应。
4.血液系统反应:白血球减少粒性白血球缺乏症，血小板减少溶血性贫血，再生障碍性贫血等反应在服用磺酰脲类降糖药时均有报道。
5.其它反应:有报道说服用格列本脲和其它磺酰脲类降糖药在饮食和视力方面有变化，这是因为血糖波动所致。此外还有关节痛、肌肉痛、血管炎和甲状腺功能低下等反应。
禁忌
1.对本品或其它磺酰脲类药物过敏者。
2.糖尿病酮症酸中毒或糖尿病昏迷者。
3.胰岛素依赖性(Ⅰ型)糖尿病患者。
4.严重肝、肾、甲状腺疾患者。
5.妊娠者。
注意事项
1.据报道，本品可增加心血管疾病的死亡危险，故须特别注意。
2.禁忌超剂量服用。
3.病人应坚持控制饮食，坚持体育锻炼，定期进行血糖或尿糖检查。
4.老年人、体质衰弱和营养不良的病人应慎用。注意初始和维持剂量，以免造成低血糖。
