适应症
镇吐药。 ①用于化疗、放疗、手术、颅脑损伤、脑外伤后遗症、海空作业以及药物引起的呕吐； ②用于急性胃肠炎、胆道胰腺、尿毒症等各种疾患之恶心、呕吐症状的对症治疗； ③用于诊断性十二指肠插管前用，有助于顺利插管；胃肠钡剂X线检查，可减轻恶心、呕吐反应，促进钡剂通过。
用法用量
肌内或静脉注射。成人，一次10～20mg（1支～2支），一日剂量不超过0.5mg/kg；小儿，6岁以下每次0.1mg／kg，6～14岁一次2.5～5mg(0.25支～0.5支)。肾功能不全者，剂量减半。
不良反应
①较常见的不良反应为：昏睡、烦燥不安、疲怠无力； ②少见的反应有：乳腺肿痛、恶心、便秘、皮疹、腹泻、睡眠障碍、眩晕、严重口渴、头痛、容易激动； ③用药期间出现乳汁增多，由于催乳素的刺激所致； ④注射给药可引起直立性低血压； ⑤大剂量长期应用可能因阻断多巴胺受体，使胆碱能受体相对亢进而导致锥体外系反应（特别是年轻人），可出现肌震颤、发音困难、共济失调等，可用苯海索等抗胆碱药物治疗。
禁忌
（1）下列情况禁用：
①对普鲁卡因或普鲁卡因胺过敏者；
②癫痫发作的频率与严重性均可因用药而增加；
③胃肠道出血、机械性肠梗阻或穿孔，可因用药使胃肠道的动力增加，病情加重；
④嗜铬细胞瘤可因用药出现高血压危象；
⑤不能用于因行化疗和放疗而呕吐的乳癌患者。
(2)下列情况慎用：
①肝功能衰竭时，丧失了与蛋白结合的能力；
②肾衰，即重症慢性肾功能衰竭使锥体外系反应危险性增加，用量应减少。
注意事项
①对晕动病所致呕吐无效； ②醛固酮与血清催乳素浓度可因甲氧氯普胺的使用而升高； ③严重肾功能不全患者剂量至少须减少60%，这类患者容易出现锥体外系症状； ④静脉注射甲氧氯普胺须慢，1～2分钟注完，快速给药可出现燥动不安，随即进入昏睡状态； ⑤因本品可降低西咪替丁的口服生物利用度，若两药必须合用，间隔时间至少要1小时； ⑥本品遇光变成黄色或黄棕色后，毒性增高。
